On the Izumoring, a strategy of rare sugar production, aldose isomerases are used to produce aldose from ketose. Generally, aldose isomerase activity is evaluated using cysteine‐carbazole method by measuring producibility of ketose from aldose. However, this method is not appropriate method for evaluation of rare sugar producibility because it evaluates reverse reaction of production for many rare sugars. In this study, the authors found that the colorimetric method, o‐amino diphenyl‐acetic acid method (ADA method), is suitable evaluation method of aldose isomerases activity for rare sugar production. ADA method showed two digits higher detection sensitivity for aldoses such as D‐glucose, D‐mannose, and D‐allose, compared with ketoses such as D‐fructose and D‐allulose. Actually, L‐rhamnose isomerase activity was evaluated using ADA method, resulting enzyme activities which is theoretically correct were obtained.
