内容記述 | Monitored by piscicidal, antimicrobial and plant growth inhibitory activities, biologically active substances were isolated from the leaves of Platycarya strobilacea SIEB. et ZUCC. Five kg (fresh weight) of the leaves were extracted with methanol, the methanol extract was partitioned with ethyl acetate and water. The ethyl acetate soluble fraction which showed the activities was fractionated by column and preparative thin layer chromatography, succesively and finally purified by sublimation to give active substances I (346 mg), II (89 mg) and III (131 mg). Active substance I, C10H6O3 was identified as 5-hydroxy-1, 4-naphthoquinone. Active substances II and III have the same molecular formula, C11H8O4. The former was identified as 5-hydroxy-2-methoxy-1, 4-naphthoquinone and the latter as 5-hydroxy-3-methoxy-1, 4-naphthoquinone. The piscicidal activity against the killie-fish of active substances I, II and III were represented by LC50 (24 hr) values of 0.56, 0.75 and 0.56 ug/ml, respectively. The active substances exhibited microbial activity against Bacillus subtilis, Escherichia coli and Saccharomyces cerevisiae, especially substance II showed the strongest activity. To Staphylococcus aureus, substance I showed strong activity, while III had no effect. Substances II and III had weak effect on Rhizopus japonicus and Aspegillus niger. Substance II showed the strongest inhibition on growth of rice seedlings among the three substances. | |||||||||||||||||

内容記述 | ノグルミの生葉5Kgをメタノールで抽出濃縮し, この濃縮液を酢酸エルと水で分配させると, 魚毒性, 抗菌性, 植物生長阻害性は酢酸エチル層へ移行した. この酢酸エチル可溶物(76.4g)をカラムクロマトグラフィーで精製し, 更に分取TLC, 昇華法により, 活性物質I(346mg), II(89mg), III(131mg)を単離した. 活性物質Iの分子式はC10H6O3で, 5-hydroxy-1,4-naphthoquinoneと同定した. IIとIIIは同一の分子式, C11H8O4を有する異性体で, 前者は 5-hydroxy-2-methoxy-1,4-naphthoquinone, 後者は5-hydroxy-3-methoxy-1,4-naphthoquinoneと同定した. 活性物質I, II, IIIのヒメダカに対する殺魚効果LC50(24hr)はそれぞれ, 0.56, 0.75, 0.56μg/mlであった. 活性物質I, II, IIIはBacillus sebtilis, Escherichia coli, Saccharomyces cerevisiaeに対して抗菌性を示し, この作用はIIが最も強かったが, Staphylococcus aureusに対してはIが強く, IIIにはその作用がなかった. また, II, IIIはRhizopus japonicus, Aspergillus nigerに対しても弱い抗菌性があった. イネ幼苗に対する植物生長阻害作用は活性物質IIが最も強かった. | |||||||||||||||||
