| 内容記述 | An α-L-arabinofuranosidase (EC 3.2.1.55) from the culture fluid of Streptomyces massasporeus grown on beet arabinan as a carbon source has been highly purified. The purified enzyme has a pH optimum of 5.0, and was demonstrated to be highly active on ρ-nitrophenyl α-L-arabinofuranoside. The enzyme was potently inhibited by Hg2+ or L-arabonic γ-lactone. The apparent Km values of the enzyme for ρ-nitrophenyl-α-L-arabinofuranoside and arabinan were determined to be 1.67 mM and 25 g per liter, and Vma values were calculated to be 8.33 and 4.88 μ moles per min per mg of protein, respectively. The molecular weight of the enzyme was estimated to be 54,000. | |||||||||||||||||||||||||||||

| 内容記述 | Streptomyces massasporeusはビートアラビナンを炭素源とする培養により, α-L-アラビノフラノシダーゼ(EC 3.2.1.55)を生産し, その酵素はカラムクロマトグラフィーにより高度に精製された. 精製酵素は分子量54,000で, pH5.0に至適pHを持ち, Hg2+およびL-アラビノ-γ-ラクトンにより活性が阻害された. Ρ-ニトロフェニル α-L-アラビノフラノシドに高い活性を示し, ビートアラビナンにも作用した. この両基質に対するKm値はそれぞれ1.67mMおよび25g/lであり, Vmaxは8.33および4.88μmol/min/mgであった. | |||||||||||||||||||||||||||||
