| 内容記述 | We recommend, as the most simple one, the following method of preparation of the fiber-sections by a new technique which is developed by us. To begin with, take a piece of cork stopper 1cm in length and parallel to its axis make an incision of about 3m depth with a razor. Next, insert a small qantity of sample into the cut and disintend its both ends with tention. By chopping with the razor, make a pillar-like piece having the dimmensions of about 5?6 mm square and about 1cm height, out of the cork holding the sample fiber in the heart. Dip the cork piece in melting paraffin just for a moment by way of tweezers and let the liquid permeat the cut. Shave the cork off with a razor or safty-razor blade as thin as possible. Mount the cork slice upon a slide-glass and examine the fiber-sections under microscope as usual. This very application of paraffin prevents the segregation of fiber-pieces from the cork-slice during the shaving work and at the same time enables us to obtain very thin sections of 30 microns range. Opaque materials such as semi-dull rayon are fairly viewed as well in slices ot about 50 microns in thickness. | |||||||||||||||||
